Study Information

Summary / Purpose

To determine whether dose-escalated radiation therapy improves 3-year overall survival compared to standard care in patients with locally advanced, unresectable pancreatic cancer who have responded to initial chemotherapy.

Study Type: Interventional

Accepts Healthy Volunteers: No

Who Can Participate

Eligibility Criteria:

Adults 18+ years of age
Diagnosed with pancreatic ductal adenocarcinoma
Locally advanced, unresectable disease (cannot be removed by surgery)
Must have completed 4–6 months of first-line chemotherapy (FOLFIRINOX, NALIRIFOX, or gemcitabine/nab-paclitaxel)
No radiographic progression and biochemical response (CA19-9 criteria) after chemotherapy
Adequate liver function and no active gastrointestinal ulceration or tumor invasion of stomach/bowel
No recent cardiac hospitalization or heart failure

What's Involved

Participation in the study will include:

Standard care (additional chemotherapy, observation, or standard-dose radiation) or dose-escalated radiation therapy delivered daily, every other day, or twice weekly over 5 or 25 fractions
Possible additional chemotherapy after radiation to complete 6 months total
CT or MRI scans to monitor response to treatment
Blood tests and tumor biopsies for monitoring
